Salmonella Enterica LT2 strain fadE (Acyl-coenzyme A dehydrogenase)
This sequence encodes a specific enzyme from the Salmonella Beta-Oxidation cycle. fadB is one of many enzymes under the fad family, which largely represents enzymes associated with fatty acid metabolism. In the Beta Oxidation cycles of Salmonella and E.coli, fadB acts as the large subunit in the tetramer fadBA, or tri-functional enzyme complex fadBA. The tetramer consists of two large subunits (fadB) and two small subunits (fadA). The fadBA complex is a part of the aerobic respiration cycle in Salmonella. It is shown in literature to be less sterically specific compared to that of E.coli (https://www.ncbi.nlm.nih.gov/pmc/articles/PMC1347308/)
The Beta-Oxidation in E.coli can be broken down into four main parts:
1)acyl-CoA dehydrogenase (ACD) 2) 2-enoyl-CoA hydratase (ECH) 3)3-hydroxyacyl-CoA dehydrogenase (HACD) 4)3-ketoacyl-CoA thiolase (KACT)
fadE is responsible for the first reactions. The activity of this enzyme can be explored here: (https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9329746/)
This part should always be used in conjunction with the fadBA genes of Salmonella Enterica. These three genes represent the complete beta-oxidation cycle of Salmonella Enterica. It should be used with the specificity of beta-oxidation in mind, and it only acts upon endogenous fatty acids. fadE recouples the biproducts of the breakdown of fatty acids, and either continues to break down the fatty acid into acetyl-coA, or feeds the long-chain fatty acid into the membrane repair cycle.
